The gig takes place at the MEN Arena on November 17, and will also feature Ed Sheeran, JLS, Elbow, N-Dubz, Hugh Laurie, Michael Buble, James Morrison and Snow Patrol.
“We've got some enormous acts, Lady Gaga, Coldplay, Snow Patrol, Michael Buble, all on the same stage I think it's just going to be an incredible evening,” Barlow told BBC Radio 1 Newsbeat.
“It's going to be the biggest music event of 2011.”
The Take That star, who organised the gig, said he had put a hit list together and received confirmation from 80% of the artists he targeted.
“It's exciting when you get a yes and start imagining how the night will come together,” he said.
“I'm really pleased everyone has come through. For anyone who buys a tickets, I think it's going to be an incredible experience.”
