The 38-year-old hip-hop artist, real name Nasir bin Olu Dara Jones, was a friend of the late singer, who died in July, at the age of 27.
Nas confirms speculation that the track was written about him in an interview with US magazine XXL, in which he also reveals that he originally met Winehouse through music producer Salaam Remi, who worked with her on her albums Frank and Back To Black.
The 2006 song Me And Mr Jones is about a man, who apparently let her down and is peppered with references that suggest it was about Nas.
Winehouse challenges 'Mr Jones' by singing: 'What kind of f***ery is this? You made me miss the Slick Rick gig/You thought I didn't love you when I did/Can't believe you played me out like that.'
Apart from the title, there are other clues that the song was about Nas.
The line, 'Mr Destiny, nine and 14', is believed to be a reference to the rapper's daughter Destiny, while “nine and 14' supposedly refers to 14 September – the birthday that Nas and Winehouse shared.
The New York-born star – who was married to R&B singer Kelis – did not indicate whether or not he had been romantically involved with Winehouse, but he did reveal that they had been planning to celebrate their joint birthday together last month, had she lived.
In August, he tweeted: “Amy was my sister. [A] genuine person who spoke her mind and didn’t give a f***.
“We shared the same [birthday]! We was [sic] planning a party this September 14th! I gotta celebrate for her!”
