The 30-year-old plays Sister Rosemary alongside Jane Lynch, who stars as Mother Superior, in the new adaptation, which is due to be released next year.
The Dreamgirls star says the movie has a lot of sentimental meaning as her mother Darnell, who was killed in 2008, liked the Stooges a lot.
She told Ebony magazine: 'It's so different from anything I've done. I play a nun. I feel like I'm getting my feet wet in comedy. Another reason that I took this role is because my mum was a huge fan of the Three Stooges.'
Jennifer's mother and her brother Jason were gunned down in their Chicago, Illinois home in October 2008.
