The US star turned up unannounced at the Comedy Store in Piccadilly Circus for an impromptu set.
The show had been billed as a normal Wednesday night and comedians such as Neil Mullarkey, Paul Merton and Josie Lawrence took to the stage for around an hour before compere Andy Smart introduced Seinfeld at 10pm.
Smart later wrote on Twitter: 'I never thought I'd ever say this, but I've just introduced Jerry Seinfeld at the Comedy Store. A good day at work!'
According to NME, almost all the audience were unaware of Seinfeld's addition to the bill and during his set he made comments about Twitter, the Royal Wedding as well as Arnold Schwarzenegger before audience members asked him questions.
Tickets for his sold-out gig at the O2 Arena on June 3 ranged from between £70 to £100.
